A Study on the Detection of Neuroblastoma Cells from Bone Marrow Using Monoclonal Antibodies and Immunobeads


Abstract
For the detection of neuroblastoma cells from bone marrow, the author used four monoclonal antibodies(459, 390, HSAN 1.2 and RB 21-7) specific for neu-roblastoma cells and immunobeads(Dynabeads).
Two clinically suspected neuroblastoma patients were studied, and as study controls one healthy child :with: acute lymphoblastic leukemia. and those -two pa-tients without adding monoclonal antibodies were used.
The following results were obtained:
Cells coated with five or more beads were not observed in the control groups.
Cells coated with one to two beads observed in two patients were not signifi-cant, compared to the control groups.
Cells coated with three to four beads observed in two patients were statisti-cally significant, compared to the control groups(p<0.01 and p<0.05).
Cells coated with five or more beads were observed only in two patients¢¥ ce-lls treated with monoclonal antibodies and immunobeads.
There were no differences according to the amount of monoclonal antibodies (15?g and 30?g/5 X 105 cells) and the number of immunobeads (2X106 and 3 X 106/5 X 105 cells).
Neuroblast-like cells were observed from the fourth day of bone marrow cell culture in Leibovitz¢¥s L-15 (with 17% Fetal Bovine Serum) media of the two patients. This finding confirmed that the observed cells coated with¢¥ immuno-beads were neuroblastoma cells.
This diagnostic procedure is a relatively simple and rapid, and may be of va-lue for the diagnosis of neuroblastoma metastasis to the bone marrow.
